Ordering Recommendation: Use to subtype deficiency in known protein S-deficient individuals in combination with Protein S Antigen (Free) [PSAGF] or Protein S, Functional.

Patient Preparation: Do not order if individual has been on warfarin therapy in the previous 2-4 weeks.

Interferences and Limitations

Cautions: Patients on warfarin may have decreased protein S values. Patients should be off warfarin therapy for two weeks for accurate measurement of protein S.

Handling Instructions

Outside Laboratory:

  • Centrifuge for 10 minutes and remove plasma.
  • Centrifuge the plasma again for another 10 minutes.
  • Transfer 1.5 mL platelet-poor plasma (1 mL minimum) into a separate plastic aliquot. Freeze plasma at -20°C to -80°C while awaiting shipment. Transport on dry ice.

Critical Frozen: Submit separate specimens when ordering multiple tests.

Stability: Frozen (-20°C): 3 months; Frozen (-70°C): 6 months; Refrigerated: Unacceptable; Ambient: 4 hours.

Unacceptable Conditions: Serum, EDTA plasma, clotted or hemolyzed specimens.

Processing

UW-MT SPS: Immediately forward specimens to Coagulation Lab for processing.
UW-MT Coag: Double-spin the sample and aliquot 2 mL platelet-poor plasma (1 mL minimum) into a separate plastic aliquot. Freeze plasma at -20°C (place in the UW Sendouts frozen rack).

HMC SPS: Immediately forward specimens to Coagulation Lab for processing.
HMC Coag: Double-spin the sample and aliquot 2 mL platelet-poor plasma (1 mL minimum) into a separate plastic aliquot. Freeze plasma at -20°C. Forward to UW-MT for sendout.

UW-NW or Fred Hutch: Centrifuge for 10 minutes and remove plasma. Centrifuge the plasma again for another 10 minutes. Transfer 1.5 mL (1 mL minimum) platelet-poor plasma into a separate plastic aliquot. Freeze plasma at -20°C. Forward to UW-MT for sendout.
