"The role of PS1/gamma-secretase in the pathophysiology of Alzheimer’s disease”
Presenilin (PS)/γ-secretase is widely recognized as the protease responsible for the processing of APP and thus for production of neurotoxic β-amyloid (Aβ) peptides in Alzheimer’s disease (AD). However, it also cleaves ~150 other substrates and has multiple interacting partners, thereby influencing numerous cellular signaling pathways that could contribute to AD pathogenesis. This may partially explain why Aβ-targeting therapies failed in clinical trials. This seminar will review “the other side” of the PS/gamma-secretase. We will also discuss molecular imaging approaches and tools, and how these may be helpful in developing new therapeutic strategies for neurodegenerative disorders.
