Description
Mycobcaterium chimaera DNA detection by PCR is only available upon request as a reflexive test if a 3-way split ID of “Mycobacterium chimaera or Mycobacterium intracellularae or Mycobacterium paraintracellularae” is reported from tissue/fluid or a pure culture.
Results in 2-3 business days after reflex.

Acceptable Specimens
For tissue/fluid acceptable specimens please refer to corresponding battery that this test can be reflexed from. Each of the following batteries have a variation in acceptable specimens.
- Nontuberculous Mycobacteria DNA Detection [NTMDNA]
- Mycobacterium avium complex DNA Detection [MAVDNA]
- Bacterial PCR reflex NGS [BCTDNA]
Pure Culture
- Plate/slant: should have visible growth
- Broth/positive blood bottles: must have organisms visible by stain to be acceptable for sequence analysis
- A mixed culture will be subject to an additional charge

Shipping/Handling
- Fresh frozen tissue/fluid specimens should be collected into a DNA free container labeled with at least two identifiers and be submitted and maintained on dry ice.
- Formalin Fixed Paraffin-embedded tissues (FFPE, PET) can be sent ambient or with ice packs during warmer summer months to prevent melting.
- Pure cultures* should be sent at room or refrigerated temperature. Follow approved regulations for transporting infectious substances.

Quantity
requested: See Optimal Quantity above
minimum: Specimens below optimal quantity are acceptable for testing, however, diagnostic yield is generally proportional to specimen size.
Processing
UWMC/HMC: Store and send fresh tissue/fluid specimens refrigerated, if specimen storage and transport will exceed 8 hours, freeze at -20°C. Freeze all fresh tissue/fluid specimens at -20°C upon arrival in UW Molecular Microbiology.
